In 2014, ibrutinib was introduced for relapsed/refractory chronic lymphocytic leukemia patients. Data from 315 patients in the UK/Ireland with a median follow-up of 16 months showed a 1-year discontinuation-free survival of 73.7% and an overall survival of 83.8%. Patients with better pre-treatment performance status had higher survival rates. Multivariable analysis revealed that older patients with 17p deletion had poorer outcomes when treated beyond the second line. While dose reductions alone did not significantly impact survival, extended treatment breaks, especially permanent cessations, were associated with reduced survival and increased mortality. The study's outcomes were lower compared to the RESONATE trial, possibly due to inclusion criteria and treatment discontinuations.
